Poelzig's house design is a well known landmark in set design, and it really adds a lot to the film, which is one of my favorites, but as some have already mentioned, Hill House was integral to the film, a real presence, so I have to vote for it.

Skull Island - to be fair it really was a toss up with Black Lagoon (Julie Adams had the best legs - hee hee) but nothing terrified me like Kong shaking those men off that tree.

The Evil Dead Cabin. I have to go with this one because it is a classic that has literally become a horror movie cliche because of this film. It was also an effectively creepy/eerie setting in the original film. Haddonfield was certainly close, in my opinion though.
